If we could expect improvement of β-cell dysfunction by reducing glucotoxicity through glucose control, it could be very helpful for diabetes management to control lipotoxicity within pancreatic islets. Seok et al. showed the anti-diabetic effect of balsamic vinegar through improvement of β-cell function via increasing β-cell ATP-binding cassette transporter subfamily A member 1 (ABCA1) expression in Otsuka Long-Evans Tokushima Fatty (OLETF) rats fed a high-fat diet. Reducing glucose levels as measured by an oral glucose tolerance test can be associated with the improvement of β-cell function. A reduction in β-cell mass can be a main cause for the development of type 2 diabetes [5,6]. Because, not only is there an increase in insulin resistance observed in OLETF rats with type 2 diabetes, there is also a severe β-cell deficit. It would be of interest to researchers and clinicians to observe changes in β-cell mass in OLETF rats fed with balsamic vinegar. In other words, there is a need to demonstrate whether balsamic vinegar increased insulin secretion from pancreatic β-cells or was there also an increase in β-cell mass. The authors showed that the insulin/islet area in pancreatic sections were stained with antibodies against insulin, however, the results would be stronger if the authors provided evidence showing the results of a β-cell mass analysis. In the study, fasting glucose and postprandial glucose levels decreased as measured by a glucose tolerance test, but the authors could not give evidence for whether this change could be associated with insulin secretion at fasting or postprandial status or due to another mechanism such as an increase in glucose uptake through the improvement of peripheral insulin resistance. It is possible that β-cell dysfunction improved through a reduction in pancreatic cholesterol staining. It could give more evidence to show quantitative analysis for the pancreatic cholesterol. Nevertheless, this study provides the meaningful conclusion that balsamic vinegar can improve lipotoxicity in pancreatic β-cells. In the future, how such a substance could suppress lipotoxicity in β-cells and how it could affect β-cell mass or dysfunction will need to be demonstrated and elucidated in greater detail.

Balsamic Vinegar Improves High Fat-Induced Beta Cell Dysfunction via Beta Cell ABCA1

BACKGROUND The aim of this study was to investigate the effects of balsamic vinegar on β-cell dysfunction. METHODS In this study, 28-week-old Otsuka Long-Evans Tokushima Fatty (OLETF) rats were fed a normal chow diet or a high-fat diet (HFD) and were provided with tap water or dilute balsamic vinegar for 4 weeks. Oral glucose tolerance tests and histopathological analyses were performed there...

Beta Cell Protective Effects of Sodium Tungstate in Streptozotocin-Induced Diabetic Rats: Glycemic Control, Blockage of Oxidative Stress and Beta Cell Histochemistry

Background: Diabetes is a major public health problem. The development of new therapies that are able to improve glycemia management and even to cure diabetes is of great interest. In this study, protective effects of sodium tungstate against STZ-induced beta-cell damages were investigated. Methods: Sixty rats were divided into six groups: control, diabetic, sodium tungstate treated diabetic r...

The Class I Histone Deacetylase Inhibitor MS-275 Prevents Pancreatic Beta Cell Death Induced by Palmitate

Elevation of the dietary saturated fatty acid palmitate contributes to the reduction of functional beta cell mass in the pathogenesis of type 2 diabetes. The diabetogenic effect of palmitate is achieved by increasing beta cell death through induction of the endoplasmic reticulum (ER) stress markers including activating transcription factor 3 (Atf3) and CAAT/enhancer-binding protein homologous p...

Carriers of Loss-of-Function Mutations in ABCA1 Display Pancreatic β-Cell Dysfunction

OBJECTIVE Abnormal cellular cholesterol handling in islets may contribute to beta-cell dysfunction in type 2 diabetes. beta-Cell deficiency for the ATP binding cassette transporter A1 (ABCA1), which mediates the efflux of cellular cholesterol, leads to altered intracellular cholesterol homeostasis and impaired insulin secretion in mice. We aimed to assess the impact of ABCA1 dysfunction on gluc...
